Protein phosphorylation is a reversible post-translational modification of proteins in which an amino acid residue is phosphorylated by a protein kinase by the addition of a covalently bound phosphate group. Phosphorylation alters the structural conformation of a protein, causing it to become activated, deactivated, or otherwise modifying its function. Approximately 13,000 human proteins have sites that are phosphorylated. The reverse reaction of phosphorylation is called dephosphorylation, and is catalyzed by protein phosphatases. Protein kinases and phosphatases work independently and in a balance to regulate the function of proteins. The amino acids most commonly phosphorylated are serine, threonine, tyrosine, and histidine. These phosphorylations play important and well-characterized roles in signaling pathways and metabolism. However, other amino acids can also be phosphorylated post-translationally, including arginine, lysine, aspartic acid, glutamic acid and cysteine, and these phosphorylated amino acids have been identified to be present in human cell extracts and fixed human cells using a combination of antibody-based analysis (for pHis) and mass spectrometry (for all other amino acids). R-30490 (also known as 4-methoxymethylfentanyl) is an opioid analgesic related to the highly potent animal tranquilizer carfentanil, and with only slightly lower potency. It was first synthesised by a team of chemists at Janssen Pharmaceutica led by Paul Janssen, who were investigating the structure-activity relationships of the fentanyl family of drugs. R-30490 was found to be the most selective agonist for the μ-opioid receptor out of all the fentanyl analogues tested, but it has never been introduced for medical use in humans, although the closely related drug sufentanil is widely used for analgesia and anesthesia during major surgery. Side effects of fentanyl analogs are similar to those of fentanyl itself, which include itching, nausea and potentially serious respiratory depression, which can be life-threatening. Fentanyl analogs have killed hundreds of people throughout Europe and the former Soviet republics since the most recent resurgence in use began in Estonia in the early 2000s, and novel derivatives continue to appear.
Based on animal research, it was initially thought that bicalutamide was unable to cross the blood–brain barrier into the central nervous system and hence would be a peripherally-selective antiandrogen in humans. This conclusion was drawn from the finding that bicalutamide reportedly does not increase LH or testosterone levels in multiple tested animal species, including rats and dogs. AR antagonists like flutamide normally do this by blocking ARs in the pituitary gland and hypothalamus in the brain and thereby disinhibiting the HPG axis. The notion that bicalutamide does not cross the blood–brain barrier is in accordance with rodent tissue distribution studies, which have found low levels of bicalutamide in the hypothalamus and cerebral cortex relative to most peripheral tissues and the pituitary gland (a part of the brain that is outside of the blood–brain barrier). In spite of the preceding studies however, other rodent studies have contradicted these results and found that bicalutamide does significantly and dose-dependently increase LH and testosterone levels, and in some studies to a similar extent as flutamide. In any case, in humans, bicalutamide has consistently been found to increase LH and testosterone levels, and to a comparable extent relative to flutamide and nilutamide. This occurs to a significant extent at even a very low dosage of 10 mg/day bicalutamide.
The meteorite contained a mixture of left-handed and right-handed amino acids; most amino acids used by living organisms are left-handed in chirality, and most sugars used are right-handed. A team of chemists in Sweden demonstrated in 2005 that this homochirality could have been triggered or catalyzed by the action of a left-handed amino acid such as proline. Several lines of evidence indicate that the interior portions of well-preserved fragments from Murchison are pristine. A 2010 study using high resolution analytical tools including spectroscopy, identified 14,000 molecular compounds, including 70 amino acids, in a sample of the meteorite. The limited scope of the analysis by mass spectrometry provides for a potential 50,000 or more unique molecular compositions, with the team estimating the possibility of millions of distinct organic compounds in the meteorite. In November 2019, along with the Northwest Africa 801 meteorite it was the first to provide evidence of pentoses (including ribose) in space, using gas chromatography–mass spectrometry. All the straight-chain five-carbon aldoses were found but tetroses, sugar alcohols, sugar acids, and deoxyribose were not detected.
