Measured responses usually involve growth hormone and insulin-like growth factor 1, known as IGF-1. Growth hormone rises in bursts and is difficult to sample reliably, while IGF-1 shifts more slowly and can be assessed from a single blood draw. Studies therefore treat IGF-1 as the more practical pharmacodynamic marker. Both are indirect, showing that the receptor was engaged rather than that the peptide reached a particular concentration. Direct exposure measurement requires an assay aimed at the molecule itself.
Published work tends to frame tesamorelin as a tool for studying the GHRH axis and as a compound with measurable effects on body composition. Reports often describe visceral adipose tissue as an endpoint, assessed by imaging rather than by inference. Analytical sections commonly describe liquid chromatography with tandem mass spectrometry to confirm identity and purity, because immunoassays may cross-react with related fragments. Where results diverge between studies, differences in assay choice, sampling timing, and population are frequent explanations offered. Whether effects persist after treatment stops remains an open question.
Tesamorelin binds the growth hormone–releasing hormone receptor on pituitary somatotroph cells. The receptor signals through the Gs protein, raising intracellular cAMP and activating protein kinase A. That cascade triggers release of stored growth hormone in pulses rather than a steady stream. Because the drug acts at the receptor that normally controls this process, its effect depends on the body's own signaling architecture rather than on a synthetic pathway. The resulting hormone profile reflects the timing of each pulse, not only its size.
Clinical study of tesamorelin has centered on adults with HIV-associated lipodystrophy, a condition in which abdominal fat accumulates while peripheral fat is lost. In controlled trials, treated participants showed reductions in visceral adipose tissue measured by imaging, alongside modest shifts in some lipid values. Effects on subcutaneous fat were smaller and less consistent across studies. Whether these changes translate into fewer cardiovascular events remains an open question, because the trials were not designed or powered to answer it.
Tesamorelin is a synthetic peptide that acts as an analog of growth hormone-releasing hormone, a natural hypothalamic signal. Its sequence corresponds to the forty-four amino acid form of the human hormone, with a small acyl group attached near the amino terminus. That modification slows enzymatic breakdown and extends the time the peptide remains active in circulation. The compound was developed as a pharmacological way to raise endogenous growth hormone output rather than supplying the hormone directly.
After injection, the peptide binds receptors on somatotroph cells in the anterior pituitary. Receptor activation raises intracellular cyclic AMP and triggers release of stored growth hormone into the bloodstream. Because the compound works through the body's own regulatory system, growth hormone pulses retain much of their normal feedback control. Repeated administration also raises insulin-like growth factor 1, a hormone produced mainly in the liver. Investigators treat that rise as a marker that the pituitary axis has been engaged.
| Property | Value | Notes |
|---|---|---|
| Primary target | GHRH receptor | Located on pituitary somatotroph cells |
| Signaling route | cAMP–protein kinase A | Gs-coupled receptor pathway |
| Downstream markers | Growth hormone and IGF-1 | Used as pharmacodynamic readouts |
| Common detection | LC-MS/MS | Separates intact peptide from fragments |
| Typical storage | 2–8 °C, protected from light | Applies to solid form before reconstitution |
Questions remain about how much of the observed fat reduction reflects direct GHRH-receptor signaling versus the downstream growth hormone and IGF-1 surge. It is also unclear whether the compound produces meaningful benefit in populations without lipodystrophy, since trials in cognitive impairment did not reach their stated goals. Long-term effects on glucose metabolism and on cardiovascular outcomes are not fully characterized. Published work generally describes effects on surrogate markers rather than on hard clinical endpoints, and independent replication of some findings is limited.
Tesamorelin acts on the growth hormone-releasing hormone receptor, a G-protein-coupled receptor found on somatotroph cells in the anterior pituitary. Binding triggers a rise in intracellular cyclic AMP, which in turn opens ion channels and raises calcium concentrations, leading to release of stored growth hormone into the bloodstream. Because the peptide works through the same receptor as the body's own GHRH, the resulting secretion follows a pulsatile pattern rather than a continuous elevation. The N-terminal modification slows enzymatic breakdown, so the signal persists longer than it would with the unmodified hormone.
Measuring the effect of a growth hormone-releasing hormone analogue requires markers that reflect pituitary output rather than the peptide itself. The two most frequently used are growth hormone and insulin-like growth factor 1. Growth hormone fluctuates sharply across the day and responds to sleep, stress, and meals, so isolated readings can be difficult to interpret. Insulin-like growth factor 1 changes more slowly and is often treated as the more stable integrated marker of axis activity.
Because growth hormone is released in pulses, single measurements can misrepresent overall secretion. Investigators sometimes use repeated sampling or overnight profiles to capture the pattern rather than a single value. Provocative testing, in which a stimulus is given and the response is tracked over time, offers another way to characterize the axis. Each approach carries trade-offs between sensitivity, burden on the participant, and the influence of non-target variables.

Transport theory provides an alternative interpretation of viscosity in terms of momentum transport: viscosity is the material property which characterizes momentum transport within a fluid, just as thermal conductivity characterizes heat transport, and (mass) diffusivity characterizes mass transport. This perspective is implicit in Newton's law of viscosity,

The goal of wound care is to promote an environment that allows a wound to heal as quickly as possible, with emphasis on restoring both form and function of the wounded area. Although optimal treatment strategies vary greatly depending on the specific cause, size, and age of a particular wound, there are universal principles of wound management that apply to all wounds. After a thorough evaluation is performed, all wounds should be properly irrigated and debrided. Proper cleansing of a wound is critical to prevent infection and promote re-epithelialization. Further efforts should be made to eliminate/limit any contributing factors to the wound (e.g. diabetes, pressure, etc.) and optimize the wound's healing ability (i.e. optimize nutritional status). The end goal of wound management is closure of the wound which can be achieved by primary closure, delayed primary closure, or healing by secondary intention, each of which is discussed below. Pain control is a mainstay of wound management, as wound evaluation, wound cleansing, and dressing changes can be a painful process.

=== Indonesia === The Indonesian National Armed Forces (TNI) has introduced the "TNI ration" (Indonesian: Ransum TNI) in the mid-1970s in order to standardize nutrition for soldiers in field. There are three types of ration and each daily ration consist of three menus (breakfast, lunch, and dinner), a pack of supplementary drinks, providing approximately 2,700 kcal (11,000 kJ) in total. The main course are usually meat-based meals (fish, beef, chicken, etc.) with cooked rice. The supplementary drinks are instant coffee, powdered fruit juice or vitamin supply, tea bags and powdered milk. The rations should be heated for 10–15 minutes with the included portable stove and solid fuel tablets (for canned meals), or by submerging in boiling water (for meals packed in retort pouches). All products are made in Indonesia and manufactured according to Indonesian military standard.
However, it is not universally accepted that objective well-being is a form of well-being in the strictest sense. This doubt is based on the idea that well-being is essentially a subjective phenomenon tied to a person's experience. According to this view, objective factors influence and indicate well-being but are not themselves forms of well-being. Some studies focus on either subjective or objective well-being. Others combine both perspectives to provide a more comprehensive picture and counterbalance their shortcomings. Although subjective and objective well-being often align, this is not necessarily the case. For example, a person scoring low on objective measures, such as low income and frail health, may nonetheless be subjectively happy.
== Burial methods == In many cultures, human corpses were usually buried in soil. The roots of burial as a practice reach back into the Middle Palaeolithic and coincide with the appearance of Homo neanderthalensis and Homo sapiens, in Europe and Africa respectively. As a result, burial grounds are found throughout the world. Through time, mounds of earth, temples, and caverns were used to store the dead bodies of ancestors. In modern times, the custom of burying dead people below ground, with a stone marker to indicate the burial place, is used in most cultures; although other means such as cremation are becoming more popular in the West (cremation is the norm in India and mandatory in big metropolitan areas of Japan). Some burial practices are heavily ritualized; others are simply practical.

Activation of the PI3K–AKT signaling pathway by insulin-like growth factor 1 (IGF1) or by AKT2 overexpression led to the upregulation of CUX1 and was associated with resistance to apoptosis, whereas treatment of cells with the PI3K inhibitor LY294002 decreased CUX1 expression and increased apoptosis. Whether the discrepancies in the results and conclusions from these two studies can be explained by differences in cell-types or other reasons remains to be verified. The auxiliary role of CUX1 in base excision repair has raised the possibility that its tumor suppressor function may be linked to its function in DNA repair. CUX1 knockdown delays DNA repair in multiple cell lines. Mouse embryo fibroblasts (MEFs) derived from a Cux1-/- knockout mouse exhibit increased genomic instability23, moreover Cux1+/−- heterozygous MEFs are haploinsufficient for DNA repair18. Yet, whether CUX1 hemizygosity augments the risk of cancer by increasing the frequency of mutations and/or genomic rearrangements remains to be formally tested.

=== 2021–2025: Post-Rush activities === In a January 2021 interview with Make Weird Music, Lifeson revealed that he and Lee were talking of working together on new music, "We're both eager to get back together and kind of get back into that thing that we've done since we were 14 years old that we love to do... we work really, really well together. So we'll see what happens with that." Lifeson reiterated the status of Rush and the possibility of continuing to work with Lee in a June 2021 interview with Eddie Trunk:
With two air drops under its belt, the 82nd Airborne was ready for the second "D-Day" operation in the division's history; Operation Neptune, or the amphibious assault portion of Allied invasion of Normandy. The division conducted Mission Boston, a component of the Operation Overlord plan. In preparation for the operation, the division was significantly reorganized. To ease the integration of replacement troops, rest, and refitting following the fighting in Italy, the 504th PIR did not rejoin the division for the invasion. Two new parachute infantry regiments (PIRs), the 507th and the 508th, provided it, along with the veteran 505th, a three-parachute infantry regiment punch. The 325th was also reinforced by the addition of the 3rd Battalion of the 401st GIR, bringing it up to a strength of three battalions. On 5 and 6 June these paratroopers, parachute artillery elements, and the 319th and 320th, boarded hundreds of transport planes and gliders to begin history's largest airborne assault at the time. Only Operation Market Garden later that year would be larger. During the 6 June assault, a 508th platoon leader, First lieutenant Robert P. Mathias, was the first US Army officer killed by German fire during the invasion. On 7 June, after this first wave of attack, the 325th GIR arrived by glider to provide a division reserve.

Byzantine territories fragmented into competing political entities. The crusaders crowned Baldwin I as the ruler of a new Latin Empire in Constantinople; it soon suffered a crushing defeat against the Bulgarians in 1205. It also failed to expand west or east, where three successor states had formed: the Empire of Nicaea and the Empire of Trebizond in Asia Minor, and the Despotate of Epirus on the Adriatic. The Venetians acquired many ports and islands, and the Principality of Achaea emerged in southern Greece. Trebizond lost the key port of Sinope in 1214 and thereafter played little role away from the southeastern Black Sea. For a time, Epirus appeared well placed to reclaim Constantinople from the Latins, and its ruler Theodore Doukas crowned himself emperor, but he suffered a critical defeat at the Battle of Klokotnitsa in 1230, and Epirote power waned. Nicaea, ruled by the Laskarid dynasty and drawing on both Byzantine refugees and local populations, blocked the Latins and the Seljuks of Rum from expanding east and west respectively. John III (r. 1221–1254) strengthened Nicaea militarily, economically, and diplomatically. He discouraged imports and promoted local production, and he made many diplomatic treaties, especially after Mongol armies ravaged Bulgaria and defeated Rum between 1237 and 1243. This chaos was an opportunity for John, and he fought many successful campaigns against the states disrupted by the Mongol invasions. Soon after his death, his grandson was usurped by Michael VIII, founder of the Palaiologos dynasty, who recaptured Constantinople in 1261.

It acts on the growth hormone–releasing hormone receptor, a Gs-coupled receptor found on pituitary somatotroph cells. Activation raises cAMP and prompts pulsatile hormone release.
IGF-1 reflects growth hormone activity but changes slowly and can be measured from one sample. Growth hormone itself is pulsatile, which makes single measurements hard to interpret.
The receptor pathway is well described, but how individual responses vary and what governs long-term outcomes remain open questions. Reported differences across studies are often attributed to assay and population factors.
It is a laboratory-made peptide that mimics growth hormone-releasing hormone. It prompts the pituitary gland to release growth hormone and has been studied mainly in adults with HIV-associated lipodystrophy.
