For the first fragment, the 5' overhang is complementary to the 5' overhang of the destination plasmid, while the 3' overhang of the final fragment is complementary to the 3' overhang of the destination plasmid. Such a design allows for all DNA fragments to be assembled in a one-pot reaction (where all reactants are mixed together), with all fragments arranged in the correct sequence. Successfully assembled constructs are selected by detecting the loss of function of a screening cassette that was originally in the destination plasmid. The amount of opiates can be high provided that the end product is not overly diluted during production, but the amount of diacetylmorphine and 6-monoacetylmorphine created depends on the skills of people making it and the time and conditions of acetylation. The use of the term kompot or compote refers euphemistically to kompot, a traditional, non-alcoholic drink popular in Central and Eastern European countries made by boiling fruit and spices. The so-called kompot method of making a crude opiate "cocktail" intended for injection involves stewing the pods of the opium poppy in nearly boiling water and an acid, and using the strained liquid (essentially like kompot is produced from the raw ingredients). However, in 1976 two Polish students from Gdańsk discovered a simple way of making it by extracting opiate alkaloids from poppy straw by using ion-exchange resin, acetone, ammonia water and a few other chemicals used in the last step of production. In this method, opium poppy pods and stems are boiled in water for a few hours without adding acid, and then filtered. Strongly acidic ion-exchange resin is then added. The resin is filtered and the opiates are recovered with the addition of ammonia water. The resulting liquid is evaporated using a chemical condenser, then when dry (it is then called glazura - glaze) it is acetylated using acetic anhydride in an anhydrous environment of a non-polar solvent, for example toluene, diethyl ether, chloroform, or most commonly acetone.

=== Typicals vs atypicals === Aside from reduced extrapyramidal symptoms, and with the clear exception of clozapine, it is unclear whether the atypical (second-generation) antipsychotics offer advantages over older, first generation antipsychotics. Amisulpride, olanzapine, risperidone and clozapine may be more effective but are associated with greater side effects. Typical antipsychotics have equal drop-out and symptom relapse rates to atypicals when used at low to moderate dosages. Clozapine is an effective treatment for those who respond poorly to other drugs ("treatment-resistant" or "refractory" schizophrenia), but it has the potentially serious side effect of agranulocytosis (lowered white blood cell count) in less than 4% of people. Due to bias in the research the accuracy of comparisons of atypical antipsychotics is a concern. In 2005, a US government body, the National Institute of Mental Health published the results of a major independent study (the CATIE project). No other atypical studied (risperidone, quetiapine, and ziprasidone) did better than the first-generation antipsychotic perphenazine on the measures used, nor did they produce fewer adverse effects than the typical antipsychotic perphenazine, although more patients discontinued perphenazine owing to extrapyramidal effects compared to the atypical agents (8% vs. 2% to 4%).

It is not definitively known who started the practice of serving the sausage in the bun. One of the strongest claims comes from Harry M. Stevens who was a food concessionaire. The claim is that, while working at the New York Polo Grounds in 1901, he came upon the idea of using small French rolls to hold the sausages when the waxed paper they were using ran out. A German immigrant named Feuchtwanger, from Frankfurt, in Hesse, allegedly pioneered the practice in the American Midwest; there are several versions of the story with varying details. According to one account, Feuchtwanger's wife proposed the use of a bun in 1880: Feuchtwanger sold hot dogs on the streets of St. Louis, Missouri, and provided gloves to his customers so that they could handle the sausages without burning their hands. Losing money when customers did not return the gloves, Feuchtwanger's wife suggested serving the sausages in a roll instead. In another version, Antoine Feuchtwanger, or Anton Ludwig Feuchtwanger, served sausages in rolls at the World's Fair – either at the 1904 Louisiana Purchase Exposition in St. Louis, or, earlier, at the 1893 World's Columbian Exposition, in Chicago – again, allegedly because the white gloves provided to customers to protect their hands were being kept as souvenirs. Another possible origin for serving the sausages in rolls is the pieman Charles Feltman, at Coney Island in New York City. In 1867 he had a cart made with a stove on which to boil sausages, and a compartment to keep buns in which they were served fresh.

=== Strikes on alleged drug traffickers === The US began deploying troops to the Caribbean in mid-August. By late November 2025, the US had amassed the largest military presence in the region since the Cuban Missile Crisis in October 1962, and was striking and sinking vessels allegedly operated by drug traffickers in the Caribbean and Eastern Pacific. A US official told a reporter that Hegseth's 13 November unveiling was "a formal operation naming for what the Joint Task Force Southern Spear ... and Southcom have already been doing in theater". Within days of the unveiling of the operation, cocaine-smuggling boats were captured in joint operations between the Dominican Republic and the US. As of September 2026, at least 234 people had been killed in the operation. Experts, human rights groups and international organizations including the United Nations have said the killings violate US and international law. In September 2026, UN Special Rapporteur on human rights and counterterrorism Ben Saul concluded that there were "reasonable grounds to believe that the reported 68 attacks that killed 223 people constituted crimes against humanity of murder under customary international law." The report rejected the U.S. claim of self-defence, finding that drug trafficking does not meet the threshold of an "armed attack" and that less-lethal alternatives such as maritime interception were available.

A proteose is a mixture of peptides produced by the hydrolysis of proteins. The term is somewhat archaic. A peptidergic agent (or drug) is a chemical which functions to directly modulate the peptide systems in the body or brain. An example is opioidergics, which are neuropeptidergics. A cell-penetrating peptide is a peptide able to penetrate the cell membrane.
== Overdose == Excessive doses of escitalopram usually cause relatively minor untoward effects, such as agitation and tachycardia. However, dyskinesia, hypertonia, and clonus may occur in some cases. Severe side effects of escitalopram overdose include seizures (which may be delayed), cardiovascular toxicity including QRS/QTc prolongation which can lead to arrhythmias (Torsades de pointes, ventricular fibrillation, and ventricular tachycardia), hypertension, and serotonin syndrome. Treatment of escitalopram overdoses typically involves supportive care, such as giving activated charcoal or giving benzodiazepines for seizures. Because of the risk of arrhythmias (which may be delayed) prolonged cardiac monitoring is strongly recommended.

Much like beta-oxidation, straight-chain fatty acid synthesis occurs via the six recurring reactions shown below, until the 16-carbon palmitic acid is produced. As part of supply provisions within the Comecon, Lot was allocated four Il-86s as barter for component manufacture; the airline deferred deliveries which were cancelled by 1987. The first Il-86 to be built was displayed at the Paris Salon International de l'Aéronautique in 1977.
